Não é simples fazer perguntas usando "Who is" ou "Who are", porque vai depender do seu conhecimento da resposta. Normalmente quando voce não sabe a resposta, sempre usará o singular " Who is?". A resposta será usando 'is" ou "are" se for singular ou plural.

Quando voce sabe a resposta, por exemplo, tem um lugar com varias pessoas reunidas e voce que saber quem são, sua pergunta será "Who are they?" . Sòmente nesse caso voce fará uso da pergunta no plural.
